Bacteria culture is used in the production of various food products such as cheese and yoghurt. The preferred bacteria are “grown” on suitable media in a controlled environment. It is later collected and prepared in a proper protective medium. These protective medium such as honey, skim milk, raffinose or glutamate are used to suspend cells to be freeze dried to protect the microorganisms against known freezing and drying damage. Freeze drying is a suitable technique for the preservation and long-term storage of a wide variety of microorganisms.
When it comes to the preservation and long-term storage of various types of microorganisms, freeze drying is a convenient application with most favorable results. Freeze drying is the process of removing water from frozen cell suspension by sublimation under reduced pressure. In a manufacturing production setting, the freeze dried bacteria culture result in “slabs” that look similar to styrofoam. These slabs of freeze dried bacteria culture vary in size and are stored in freezers until they are ready for use. The size of the slabs is reduced to a powder form so that they can be used in various manufacturing processes such as in the cheese fermentation process.
Every lab has different instruments; however, the following are key factors that should be considered in freeze drying bacteria culture:
- There is no specific that will work for all microorganisms. A method developed for one freeze dryer will not necessarily
translate to a different make or model.
- Not all microbes can be successfully freeze dried. Certain strains, such as mutants with deficient membranes, may not
survive the process or may die off rapidly once freeze dried.
- Lyoprotective media, such as skim milk solution, sucrose solution, or other freeze drying buffers can dramatically impact
survival rates.
- Vials used for freeze drying should always be made of glass. Atmospheric water can diffuse into plastic tubes and damage
freeze dried samples. Furthermore, samples are safest when flame sealed under vacuum in glass ampoules or tubes.
- Freeze drying protocols should be tested before committing to large scale projects. Evaluation can take 1-2 months and
yield results that indicate cells will be stable for long durations.
- The liquid used to rehydrate freeze dried bacteria can impact viability. Culture broth, e.g., TSA or LB, is often used, as
is 0.9% NaCl.
For optimum results, it is beneficial to use a gentle milling process for size reducing freeze dried bacteria culture. As well, there should be insignificant heat produced which permits maximum throughput while minimizing retention time in the mill. This helps to maximize the viability counts of the cell, That is, the number of live, unharmed cells. Using an application where the entire process directly benefits from the low heat, low energy, gentle processing, makes for a more efficient and cost effective milling process for size reducing freeze dried bacteria culture.

Proposed dates for the origin of cheese making range from around 8000 BCE (when sheep were first domesticated) to around 3000 BCE. The first cheese may have been made by people in the Middle East or by nomadic Turkic tribes in Central Asia.
Since animal skins and inflated internal organs have, since ancient times, provided storage vessels for a range of foodstuffs, it is probable that the process of cheese making was discovered accidentally by storing milk in a container made from the stomach of an animal, resulting in the milk being turned to curd and whey by the rennet from the stomach. There is a legend with variations about the discovery of cheese by an Arab trader who used this method of storing milk.
Cheese making may have begun independently of this by the pressing and salting of curdled milk to preserve it. Observation that the effect of making milk in an animal stomach gave more solid and better textured curds, may have led to the deliberate addition of rennet.
The earliest archeological evidence of cheese making has been found in Egyptian tomb murals, dating to about 2000 BCE. The earliest cheeses were likely to have been quite sour and salty, similar in texture to rustic cottage cheese or feta, a crumbly, flavourful Greek cheese.
Cheese produced in Europe, where climates are cooler than the Middle East, required less salt for preservation. With less salt and acidity, the cheese became a suitable environment for useful microbes and moulds, giving aged cheeses their respective flavours.
Ancient Greek mythology credited Aristaeus with the discovery of cheese. Homer”s Odyssey (8th century BCE) describes the Cyclops making and storing sheep”s and goats” milk cheese. From Samuel Butler”s translation ” We soon reached his cave, but he was out shepherding, so we went inside and took stock of all that we could see. His cheese racks were loaded with cheeses, and he had more lambs and kids than his pens could hold. When he had so done he sat down and milked his ewes and goats, all in due course, and then let each of them have her own young. He curdled half the milk and set it aside in wicker strainers”.
By Roman times, cheese was an everyday food and cheese making a mature art. Columella”s De Re Rustica (circa 65 CE) details a cheese making process involving rennet coagulation, pressing of the curd, salting, and aging. Pliny”s Natural History (77 CE) devotes a chapter (XI, 97) to describing the diversity of cheeses enjoyed by Romans of the early Empire. He stated that the best cheeses came from the villages near Nimes, but did not keep long and had to be eaten fresh. Cheeses of the Alps and Apennines were as remarkable for their variety then as now.
A Ligurian cheese was noted for being made mostly from sheep”s milk, and some cheeses produced nearby were stated to weigh as much as a thousand pounds each. Goats” milk cheese was a recent taste in Rome, improved over the “medicinal taste” of Gaul”s similar cheeses by smoking. Of cheeses from overseas, Pliny preferred those of Bithynia in Asia Minor.

Most men enjoy a hearty breakfast so start your Dad”s birthday off with a meal the kids can help with. Fruit fizzies, Toad in the Hole and frozen melon pops are easy and kid friendly to prepare.
Fruit fizzies are fun and frothy. Fill a glass with ice, then half full with lemon lime soda, plain carbonated soda, or sparkling mineral water. Top it off with your favorite fruit juice. Garnish with an orange slice.
The frozen melon pops should be prepared the night before so they have time to freeze. Cut watermelon, cantaloupe and honeydew melons in bite size pieces. For faster preparation, buy prepared melon chunks from the grocer”s produce case. Dip in a mixture of half lime juice and half water for flavor and to prevent discoloration. Thread the melon chunks onto a wooden skewer and freeze overnight. Serve as they are or with a dip of yogurt sweetened with honey.
Toad in the Hole is a fun dish for kids. It doesn”t take a lot of expertise to assemble and is pretty hard to wreck. Spray an oversized muffin pan with cooking spray. This is important. Cooked egg can be very difficult to remove from a baking dish. The cooking spray makes it much easier. If you don”t have the large size muffin pan, use an oven proof 8 oz glass dish. You will need a slice of bread, cooked bacon, pat of butter, and an egg for each serving. The toad is the egg and the hole is the slice of bread.
Put 1/2 a pat of butter in the bottom of each muffin tin. Gently place a slice of bread in the tin pushing it down to form a hollow. Break an egg into a measuring cup and then slide the egg from the cup into the bread hollow. Breaking the egg into the cup allows you to remove any pieces of shell. Salt and pepper to taste. Place the other half of the pat of butter on top of the egg. Crumble one slice of bacon over the top of the egg.
If your Dad really loves bacon, before you add the egg to the bread hollow, crumble a slice of bacon directly over the bread in the muffin tin, add the egg and then crumble a second piece of bacon on top. Bake in a pre-heated 350 degree oven for about 15 minutes. Check the egg to see if it”s done by jiggling it. If it”s wobbly it probably needs another minute or two of baking.
Remove the Toad in the Hole with two spoons. If Dad has a big appetite add some toaster hash browns to the plate.

Provo is a city that has a lot to offer both inhabitants and visitors. One of the strengths of the city is the dining options in the area, which are plentiful and varied in nature.
There are many different restaurants that dot the city of Provo, offering diverse dining options for those that are interested in them. There is a great combination of both unique eateries and popular chains, allowing people to pick and choose what they would like to have.

Chinese cuisine is also readily available. Indeed, there are many options available to people, from humble Chinese diners to more large scale Asian fusion locales that use Chinese food as their base for their entrees.

As with all of the other offerings, pricing and quality is variable, and can be tailored to match what people are feeling. The variety is one of the strengths of the city, and it greatly enhances the dining experience that people have.
There are also many other offerings that are more eclectic in nature, as well as numerous bistros that offer fresh foods and light fare. Essentially, consumers have the ability to find what they want when they want it and match it to their budget, making Provo a great place to eat in.
Provo is able to offer a wide range of variety that comes with the proceedings that should temp the palate of anyone that enjoys eating. Dining is always something that should bring with it feelings of satisfaction and happiness, and that is what the offerings of the city are able to achieve.
One of the great things about Provo is the varied and excellent restaurant options that dot the city. One may find any kind of cuisine that they desire, and these locations are all within close proximity of each other, making dining fun and easy.

Finding good restaurants can be a lot of fun. When traveling to a new area or if a person has recently moved, it can seem a little daunting at first. But with some planning and an adventurous spirit, it can be a pleasurable experience.
First thing to do is check a listing of eateries on the internet. A person will be able to find a listing of different cuisines, prices and reviews from previous customers. Also ask people you know if they have eaten there and if so, what was their impression. Do not go with the first suggestion, ask several people. Also check the local newspaper if you are visiting in a different area. If you are at the hotels cafe, ask the wait staff where they go for really great food.
Then before you go any further, call to make certain they are still in business. Verify their hours of service and if reservations are recommended. Was the person who answered helpful or did they sound inconvenienced by the call. Their attitude and willingness to offer help over the phone is a good indication as to the attitude of the entire staff.
If their menu is online, look at it. Look at the number of items listed and the descriptions. This is an opportunity to try something different and be a little adventurous.
Some of the restaurants will state they offer a fusion, which means they are combining different preparation styles. Others will claim to be authentic, which means they use original recipes and preparation techniques from the region of origin. Examples are authentic Italian or authentic Greek.
Another great thing about using the internet is the ability to check on the pricing of the items. People can decide if the eatery is out of their budget or just right. No matter how great the food taste and wonderful the service is, if the price is out of a person”s budget, the overall memory will not be a good one. Also, check for specials and coupons online. This will be the first visit and discounts make it all the sweeter.
When driving or walking to the restaurant, take a look at the customers. If it appears they are taking great pleasure in their meals, it is probably a place you want to try. If they are picking at their plates, you may want to reconsider. Does it appear the staff is attentive, or do you see them standing by the register or bar, chatting and snacking?
Once inside, take a whiff, good smells will equal good food. Rotten or mystery smells mean the same for the food. Then after ordering drinks, excuse yourself and go to the restroom. The cleanliness of that space will give you an excellent idea on how clean the kitchen may be.
Now that the cleanliness has been checked, the aroma is mouthwatering and the patrons seem happy, order an appetizer first. Doing so is a great way to get a sense of the food preparation abilities.
Finding good restaurants really can be a lot of fun. Be adventurous and let it be something to enjoy either alone or with others. Keep a log of places you have tried. Write your impressions and include the location of the business. People will soon be asking you where to eat and maybe even invite you to join them.

Earthquakes and terrorist attacks have been shown to disrupt the lives of many people suddenly and unexpectedly Floods and tornadoes damage peoples homes and make them unlivable Sometimes entire communities are wiped off the map and there is no place for survivors to go for meals or anything else
All of these types of events force people from their homes They usually end up in a central place like a stadium school gymnasium or other gathering place Because so many of these events are unpredictable and happen quickly people have no way to take food with them from home Depending on the scope of the disaster people may be stuck in one place for days or weeks

Vegetables are almost inconvenient to try to eat. You either have to pay extra for them with your meal if you are at a restaurant, or you have to go get them at the super market and spend half of your day preparing them.
This seems like a lot of work for many people, luckily there are easy ways to get the powerful benefits of vegies in your diet, without going out of your way. First thing, veggie smoothie to start the day, getting a good nutritious breakfast is going to change your entire day.
That is why they call it the most important meal of the day, because it really is. For my veggie smoothie, I like to get a good head of spinach and some strawberries and a banana.
The banana and strawberries are not vegetables, I know, but they do a great deal for the taste of the smoothie, and they are still very good for you. Another good ingredient for smoothies is wheat grass; this amazing grass has an astounding impact on health.
One shot of this wheat grass is going to supply your body with over 90 healthy minerals; just 1 ounce of this stuff is going to give you the equivalent benefits of eating one pound of green lettuce. That is nothing to sneeze at, and is why they call this stuff a super food.

But we need the amino acids from vegies to build and synthesize those proteins. So with your veggie smoothie in the morning put a good scoop of protein powder in there, and then go on a morning jog.
Do this for a month and you will be a changed man. These plants will also do a great deal for cleaning your skin from acne, and making you altogether a happier more well-rounded person.
